    研究工作:
  • 細(xì)胞核是一個(gè)高度組織化的三維結(jié)構(gòu),其中DNA、RNA 和蛋白質(zhì)協(xié)同作用,精確調(diào)控基因表達(dá)和細(xì)胞命運(yùn)。在生命起始階段,精子和卵子的融合形成受精卵,并發(fā)育為新個(gè)體。在此過程中,染色質(zhì)的三維結(jié)構(gòu)和空間定位經(jīng)歷劇烈重塑,這些動(dòng)態(tài)變化與基因組激活、細(xì)胞發(fā)育潛能轉(zhuǎn)換和細(xì)胞分化密切相關(guān)。

    本實(shí)驗(yàn)室致力于開發(fā)和應(yīng)用高靈敏的染色質(zhì)分析、DNA/RNA/蛋白互作檢測(cè)以及三維基因組操控技術(shù),并結(jié)合發(fā)育生物學(xué)、分子生物學(xué)、表觀遺傳學(xué)和生物信息學(xué)等多學(xué)科手段,采用干濕實(shí)驗(yàn)結(jié)合的策略,探索以下核心科學(xué)問題:

    1)哺乳動(dòng)物生殖和發(fā)育過程中染色質(zhì)三維結(jié)構(gòu)的動(dòng)態(tài)重塑及其生物學(xué)功能;

    2)RNA,特別是非編碼RNA,如何調(diào)控細(xì)胞核結(jié)構(gòu)(Nuclear organization)、基因組互作和基因表達(dá);

    3)染色質(zhì)三維結(jié)構(gòu)對(duì)人類健康和疾病的影響,并嘗試通過調(diào)控核內(nèi)三維空間結(jié)構(gòu)開發(fā)新的檢測(cè)和干預(yù)策略。

    這些研究不僅有助于深化對(duì)RNA 介導(dǎo)的染色質(zhì)調(diào)控和基因表達(dá)調(diào)節(jié)的理解,還將為改善生殖健康、提高生育力及防治胚胎發(fā)育缺陷提供理論支持和創(chuàng)新技術(shù)。
