  •   1935年2月20日生于上海市,九三學(xué)社社員。1957年9月畢業(yè)于上海復(fù)旦大學(xué)化學(xué)系。1957年至今在中國科學(xué)院上海生化細胞所進行科研(研究實習(xí)員,助理研究員,副研究員,研究員,科學(xué)院院士)。從事放射生物學(xué)、激素對基因轉(zhuǎn)錄調(diào)控,近十年來進行精子在附睪中成熟的分子機制研究。1983-1999年間在英國ICRF、美國Chicago大學(xué)、Wisconsin大學(xué)(Madison)、North Carolina大學(xué)、荷蘭ERUSMUS大學(xué)、澳大利亞Sydney大學(xué)等進行合作科研。從1993年至今,先后獲得國家自然科學(xué)三等獎和二等獎、中科院自然科學(xué)一等獎、第二屆科技十大女杰、上海市自然科學(xué)一等獎和科技功臣和何梁何利生命科學(xué)獎等。

    研究工作:
  • 主要從事雄激素對真核基因轉(zhuǎn)錄調(diào)控機制研究和睪丸生精過程中及附睪對精子成熟過程中的基因表達程序的調(diào)控,現(xiàn)集中在附睪功能基因組研究。該實驗室由張友端教授在1978年建立,她于1989年退居二線做顧問后,由其學(xué)生張永蓮教授擔任課題組長。由于這是我國第一個從事這一飛速發(fā)展的領(lǐng)域的研究室。因此他們首先建立了各種激素受體的測定方法,并以培訓(xùn)班的形式向全國推廣,以填補我國這一領(lǐng)域在研究和臨床應(yīng)用方面的空白。然后進行了雌激素受體的純化,制備單克隆抗體等基礎(chǔ)工作。同時篩選了研究激素作用原理的靶基因系統(tǒng),通過對大鼠前列腺中PSBP基因表達的時空秩序以及雄激素影響下的特征,證明了它是研究雄激素對真核基因轉(zhuǎn)錄的調(diào)控機制的一個理想系統(tǒng)。通過對PSBP的C3基因啟動子上的順式調(diào)控元件和細胞核中的反式作用因子相互作用的研究,發(fā)現(xiàn)了啟動子上的一些重要元件如:TATA盒和CCAAT盒上除了有一些通用因子與之作用外,還有組織特異的和雄激素調(diào)控相關(guān)的因子與之直接或間接地結(jié)合然后起協(xié)同作用。這些成果獲得了1993年的中國科學(xué)院的自然科學(xué)一等獎,1997年國家自然科學(xué)三等獎。為了深入了解多元件,多因子的相互作用中的調(diào)控激發(fā)程序,該課題組進而開始研究基因轉(zhuǎn)錄活性與染色體結(jié)構(gòu)的關(guān)系。

    此外,為了將基礎(chǔ)研究與生物學(xué)重大問題相結(jié)合,我們已開始研究睪丸生精過程及附睪對精子成熟過程中的基因表達程序的調(diào)控,以期能為男性健康生育調(diào)控的藥物設(shè)計提供新的思路。附睪是一根連接睪丸和輸精管的盤曲的細長管道。但不同部位的附睪細胞表達不同的基因,管腔中分泌的種類各異的蛋白質(zhì)與從睪丸中分化完全的精子相互作用,使精子表面蛋白質(zhì)的成分得以部分改變或修飾,從而使精子逐步獲得運動、獲能、精卵結(jié)合等能力而成熟,被儲存在其尾部等待射精。本組目標是克隆附睪特異表達的基因,建立一個研究精子成熟功能的測試平臺去研究這些基因在精子成熟中的作用及其表達調(diào)控。這不僅有助于揭示精子在附睪中成熟的分子機理,而且為解決精子成熟異常所引起的不孕癥的診斷和治療和開發(fā)一些阻斷精子成熟的男性避孕藥物研制提供新的藥靶和設(shè)計思路。
            目前有三方面的工作進行重點開展:
            (1) 附睪特異表達的新蛋白在精子成熟中的功能(條件性knock out/in);
            (2) 附睪特異表達的miRNA在精子成熟中的作用;
            (3) 表觀遺傳修飾(DNA methylation & histon modification)在附睪和精子成熟中的作用。

    目前正聯(lián)合所內(nèi)外力量,優(yōu)勢互補,在此基礎(chǔ)上趁勝追擊,為我國附睪與精子相關(guān)的功能基因組學(xué)和男性節(jié)育和不育作出更大的供獻。實驗室已經(jīng)和國際上的許多實驗室有合作課題,遍及美國、英國、澳大利亞和香港地區(qū)。目前,課題經(jīng)費來源國于國家自然科學(xué)基金委、中國科學(xué)院、上海市科委及美國NIH國際合作項目等。
